屋内実験の重要性

 汚濁水を生物学的浄化法を応用し、その設備の設計・維持管理を適正に実施する場合には、先ず、屋内実験により検証するする必要がある。
 また、生物学的に汚濁水を処理すると、副産物である増殖した微生物、すなわち、余剰汚泥が発生する。この余剰汚泥を効果的に処理・処分して再利用する必要がある。この汚泥の処理・処分・再利用を効率的に実施するためにも、屋内実験により処理方法を検証しなければならない。

The importance of indoor experiments

 In the case of applying the biological purification method to the polluted water and properly designing and maintaining the equipment, it is first necessary to verify it by indoor experiments.
 Further, when biologically treated polluted water is generated, a byproduct of grown microorganisms, that is, excess sludge is generated. It is necessary to effectively treat and dispose of this excess sludge for reuse. In order to efficiently carry out the treatment, disposal, and reuse of this sludge, the treatment method must be verified by indoor experiments.

屋内実験の難しさ

 公共下水道の終末処理場や事業所排水処理施設の活性汚泥を種汚泥として、屋内で活性汚泥法(回分・連続を問わず)を運転すると、SRTを数サイクル経過すると、様々な原因による汚泥膨化が発生、SVが増加し沈降分離が困難となる。
 編集者の経験では、下記(1)(2)の人工下水(水道水を利用)を用いた屋内実験(10L曝気槽 + 5L沈殿分離槽の規模)を数年の連続稼働において、糸状菌の発生や発泡現象は全く生じなかった。
 (1) 余剰汚泥を電解塩素殺傷・返送する汚泥減量型活性汚泥法
 (2) 純酸素を用いたDO 10mg/L以上で高酸素濃度型活性汚泥法(余剰汚泥なし)
 また、適正な負荷・SRT・温度・DO・pHの管理を行えば、数年間に及ぶ連続稼働において、活性汚泥法の継続運転が不可能となるような汚泥性状の悪化は認められなった。

The difficulty of indoor experiments

 When activated sludge method (regardless of batch or continuous) is operated indoors with activated sludge of public sewer terminal treatment plant or wastewater treatment facility of business establishment as seed sludge, the sludge swelling, due to various causes after several cycles of SRT, occurs, SV increases, and settling separation becomes difficult.
 According to the editor’s experience, the following indoor experiments (1) and (2) using artificial sewage (aeration tank of 10L + settling separation tank of 5L) showed no occurrence of filamentous bacteria or foaming phenomenon during continuous operation for several years.
 (1) Sludge reduction type activated sludge method for electrolytic chlorine killing and returning of excess sludge.
 (2) High oxygen concentration type activated sludge method with DO of 10 mg/L or more using pure oxygen (with no excess sludge).
 Moreover, if proper load, SRT, temperature, DO, and pH were controlled, no deterioration of sludge properties, that would make continuous operation of the activated sludge process impossible, was observed during several years of continuous operation.

 本サイトに記載の活性汚泥法(回分・連続を問わず)の屋内実験に関する照会・質問は多数ある。本ページでは、これらの照会・質問事項への回答を行った中で、実験上留意すべき共通事項について、解説する。
 環境関連(特に、生物利用によるガス・水・土壌などの浄化)に係る分野(教育研究、行政、公共施設、事業所、施設・設備・資材・薬品の設計・製造・施工・販売、委託管理など)の担当者で活性汚泥法の経験がなく、知識のみで業務を行ってトラブルを抱えているケースは少なくない。
 生物浄化に係る実務において、好気・無酸素・嫌気法に関わらず、活性汚泥法の具体的な運転管理は、様々な情報・技術を提供してくれることを強調したい。
 なお、実験上の注意事項については、未記載の事項が多数あり、順次、追加するので、更新日に留意されたい。本ページに記載する内容は、上記の総目次に示すⅠ~Ⅳのページを既読していることを前提としている。

Purpose of this page

 There are many inquiries and questions regarding indoor experiments of the activated sludge method (regardless of batch or continuous) described on this site. On this page, we will explain the common items that should be noted in the experiment while answering these inquiries and questions.
 In particular, environmental fields related to purification of gas, water, soil by biological use, there are many cases that the person does not have experience in the activated sludge method and has troubles by doing work or business only with knowledge. His or her charge is education, research, administration, public facilities, design/manufacture/construction/sales of facilities/equipment/materials/chemicals, or consignment management.
 In the practice of biological purification, we would like to emphasize that “ specific operational management of the activated sludge method provides various information and technology” regardless of aerobic, anoxic, and anaerobic methods.
 Regarding experimental precautions, there are many items that have not been described and will be added in sequence, so note the date of update. The contents on this page are based on the assumption that you have already read pages I to IV shown in the total table above.

1.初心者・実験への助言

(1)計算式に囚われないこと

 本サイトでは、活性汚泥法の計画・装置設計・維持管理に係る計算式を挙げているが、それを余り気にしないで実験されたい。
 具体的な排水量・汚濁物質の濃度から浄化装置を設計・施工・維持管理をする場合の参考として記載している。実際の条件は個々により異なるので、参考値として扱っていただきたい
 とは言え、人工下水の室内実験において、様々な困難を乗り越えていく中で、到達する実験条件は上記ページ 2)の計算結果に近づいていく。編集者自身もこの事実に驚いている。

1. Advice for beginners and experiments

(1) Don’t get trapped in the formula

 This site lists calculation formulas related to planning, equipment design, and maintenance of the activated sludge method, but please do not worry so much about them in the experiment.
 It is provided as a reference when designing, constructing, and maintaining a purification device based on specific wastewater amounts and concentrations of pollutants. Actual conditions vary depending on the individual, so treat it as a reference value.
 However, in the indoor experiment of artificial sewage, while overcoming various difficulties, the experimental condition to reach approaches the calculation result of Page 2) above. The editor himself is amazed at this fact.

(2)失敗は知識・技術の向上であること

生物を扱う実験は、物理や化学の実験と異なり、長期間の検証が必要となる。この実験期間中に様々な現象に遭遇し、その対処に知識と経験が必要となる。この課程で、多数の挫折により諦める初心者も少なくない。しかし、この課程を乗り越えることが大切で、どの分野においても、失敗経験は知識・技術の蓄積となることを認識されたい。

(2) Failure is to improve knowledge and skills

Biological experiments require long-term verification, unlike physical and chemical experiments. Various phenomena are encountered during this experimental period, and knowledge and experience are required to deal with them. In this course, many beginners give up due to many setbacks. However, it is important to overcome this course, and it should be recognized that in any field, experience of failure results in the accumulation of knowledge and technology.

2.活性汚泥の特徴

(1)開放系・混合系であること

 活性汚泥法は、開放系であるため、流入汚水・大気中に存在する様々な微生物やその胞子・種・卵などが混入し、多様な生物が共存・競争して生息する生態系である。屋内実験であっても、環境・条件によって、これらの生態系が多様に変化する。汚泥の状況が好調であったり、不調となったりする。活性汚泥が正常に機能するためには、適切な実験装置(高額な装置を意味していない)と維持管理が必要となる。
 同じ生物実験でも、厳密に管理され至適条件で行われる純粋培養系とは大きく異なる。微生物の取扱いに習熟した経験者でも、活性汚泥を適正に扱えることとは限らない。

2. Characteristics of activated sludge

(1) Open and mixed system

 Since the activated sludge method is an open system, various microorganisms, spores, seeds, eggs, etc. existing in the wastewater and the atmosphere are mixed into it, which is an ecosystem where various organisms coexist and compete. Even in indoor experiments, these ecosystems change in various ways depending on the environment and conditions. The condition of sludge is good or bad. In order for activated sludge to function properly, proper laboratory equipment (which does not mean expensive equipment) and proper maintenance are required.
 Even in the same biological experiment, it is significantly different from pure culture system, which is strictly controlled and performed under optimal conditions. Even an experienced person who is well versed in handling microorganisms cannot always handle activated sludge properly.

(2)物理・化学の実験と異なること

 化学反応などでは、再現性が良好であれば、短期間の実験で結果を確認できる。生物反応の場合には、生息環境の変化に適用できる微生物が増殖するまでに、一般的に数ヶ月を要する。また、自然界にない人工化学物質に順応できる微生物が増殖するには、数年間の連続実験が必要となることも少なくない。
 細菌類は、突然変異やウイルス等を介して、新たな環境に対応できる遺伝子を導入し、世代交代を繰り返して、その環境に順応する。これを「馴致」または「育種」という。これを早めるために、現場で稼働している活性汚泥、排水溝の底泥中の細菌類、あるいはその配管内壁に付着した生物膜を、「種」として育種することも多々ある。

(2) Different from physics/chemistry experiments

 For chemical reactions, if reproducibility is good, the results can be confirmed by short-term experiment. In the case of biological reactions, it generally takes several months for microorganisms that can adapt to changes in habitat to multiply. In addition, it is not uncommon for several years of continuous experiments to be necessary for the growth of microorganisms that can adapt to artificial chemicals not found in nature.
 Bacteria adapt to the environment by introducing genes capable of adapting to the new environment through mutations, viruses, etc. and repeating generational changes. This is called “acclimation” or “breeding“. In order to accelerate this, it is often the case that activated sludge operating in the field, bacteria in the bottom mud of the drainage ditch, or a biofilm attached to the inner wall of the pipe is bred as a “seed”.

3.活性汚泥の維持管理での留意点

(1)回分法のサイクル回数/日

 本サイトに示したバケツ(曝気容器については、後述する)による回分実験(活性汚泥法-回分実験)の件については、1 日 1 回のサイクル(静置・排出・投与、以後、翌日の静置まで連続曝気)とする。休日のときは、休日分の負荷(濃度を休日数の倍率で上げる、容量が決まっているので)で運転し、休日明けに、次回の操作を行う。粉末投与法については、後述する。
 ただし、自動制御による回分実験には、目的が異なるので、計画に沿ってサイクル回数/日で実施することとなる。回分実験では、次に示す実設備のような数サイクル/日で行う必要はない。なお、家庭用浄化槽の汚泥引抜は、1 回/年となるように設計されている。

3. Points in maintaining and managing activated sludge

(1) Batch cycles per day

 Regarding the case of the batch experiment (activated sludge method – batch experiment) by the bucket (aeration container will be described later) shown on this site, one cycle (settling/discharging/feed, then aeration until the next settling) per day. If it is a holiday, drive with the load for the holidays (the waste concentration is increased by the number of holidays and the capacity is fixed), and the next operation is performed just after the holiday. The powder feed method will be described later.
 However, since the purpose of the batch experiment with automatic control is different, it will be carried out at the number of cycles/day according to the plan. In batch experiments, it is not necessary to carry out several cycles/day like the actual equipment shown below. The sludge removal from the Jokaso (domestic septic tank in Japan) is designed to be done once a year.

実処理設備のサイクル
 現場の回分法設備では、1日に数回のサイクルで運転するが、これは、後段の余剰汚泥の処理装置の能力による。一般の下水処理場では、混合汚泥(初沈汚泥、余剰汚泥)を濃縮、(嫌気消化)、脱水・乾燥、焼却(有効利用)します。サイクル数/日を増やすのは、後段設備の安定稼働のためで、1 日 1 サイクルでは貯留・調整設備が大きくなり、不経済となるからである。
Cycle of actual processing equipment
 The on-site batch method equipment operates several cycles per day, which depends on the capacity of the drawn sludge treatment device in the subsequent stage. In general sewage treatment plants, mixed sludge (first settled sludge and excess sludge) is concentrated, (anaerobic digestion), dehydrated/dried, and incinerated (and effective use). The reason for increasing the number of cycles/day is for the stable operation of the post-stage equipment, and the storage/adjustment equipment becomes large in one cycle per day, which is uneconomical.

DO計測制御では、数 mg/L(1<DO<3) に設定する。DO制御器がない場合には、上図のように曝気量を設定する。DO上昇が生じる時点が、汚水中BODが消費される時間である。なお、汚泥抜取の時間帯は、何時でもよいが、抜取後、曝気槽の所定レベルまで水道水を補給する。

The daily cycle consists of aeration, sludge withdrawal, settling, supernatant drainage, and feed.
In DO measurement control, set to several mg/L (1 < DO < 3). If there is no DO controller, set the aeration amount as shown above. The time when DO rises is the time when BOD in the wastewater is consumed. The time for sludge extraction may be any time, but after extraction, tap water is replenished to a predetermined level in the aeration tank.

(2)汚泥日令の管理(余剰汚泥の引抜)

活性汚泥法と汚泥日令 SRT
図2に細菌類のライフサイクルを示し、表1に各種活性汚泥法の操作条件を示す。以下、標準活性汚泥法を「標準法」、長時間エアレーション法を「長時間法」と略称する。
 ○細菌の生育は、誘導期(増殖準備期間) 0~3日
 ○対数増殖期(生菌数が対数的に増加): 3~9日
 ○定常期(生菌数が一定): 10~17日
 ○衰退期(生菌数が対数的に減少): 18日以降
 標準法では、対数増殖期の初期間で増殖(生育)が極めて旺盛(基質摂取速度が最大)な細菌類を利用する。長時間法は、安定期から衰退期の細菌類(基質摂取速度は低い)を利用する。
以上のことから、運転の難易度は次のようになる。
標準法:①汚泥濃度は低い、②汚泥管理(引抜)の厳密化(適正汚泥日令の範囲が狭い)、③負荷(高負荷)/曝気バランスの厳密化(DO制御)、④汚泥状態が不安定となりやすい、⑤設置面積が狭い、⑥高度の管理技術 ⇨ 都市部の大型下水処理場
長時間法:①汚泥濃度が高い、②汚泥管理(引抜)の容易化(適正汚泥日令の範囲が広い)、③負荷(低負荷)/曝気のバランスの容易化、④汚泥状態が安定している、⑤設置面積が広い、⑥高い管理技術が不要 ⇨ 小規模下水処理場、中型・大型浄化槽(集落排水を含む)
 以上のことから、初心者が室内実験をするときには、先ず、長時間法をマスターする必要があろう。
汚泥日令 SRT の管理
 室内実験では、小容量の曝気槽となるので、汚泥日令の管理は、曝気槽内の混合液を毎日、抜き取る。沈殿槽の返送汚泥(汚泥濃度が一定しないので)を抜き取ることはしない。
 有効容量10 Lとした場合には、曝気・攪拌状態で、混合液を 1 L 余剰汚泥として抜き取ると、SRT は 10 日となる。0.5 Lであれば、20 日となる。n 日の間隔の抜取であれば、SRT = 10/nX として nX [L]の余剰汚泥を引き抜く。引抜間隔は毎日でも数日でもOKである。
 SRT は、標準法は 2~5日、長時間法では 15~30日、MLSS は前者で 1,500~2,000 mg/L、後者では 3,000~6,000 mg/L の範囲であればよい。汚泥管理は、混合液の抜取のみとし、MLSS は自然値に任せ、特に調整する必要はない。標準法については、容積負荷と汚泥日例の厳密な管理が必要である。

(2) Management of sludge days (extraction of excess sludge)

Fig.2 below shows the life cycle of bacteria, and Table 1 below shows the operating conditions for various activated sludge processes. Hereinafter, the standard activated sludge method is abbreviated as “standard method” and the long-time aeration method is abbreviated as “long-time method”.
 ○ Bacterial growth is 0 to 3 days during induction period (proliferation preparation period).
 ○ Logarithmic growth phase (the number of viable bacteria increases logarithmically): 3 to 9 days.
 ○ Stationary phase (constant viable cell count): 10 to 17 days.
 ○ Decline (live log count decreases logarithmically): After 18 days.
 The standard method utilizes bacteria that are extremely active in growth (growth) (maximum substrate uptake rate) during the early stages of the logarithmic growth phase. The long-term method utilizes bacteria from the stable phase to the declining phase (low substrate uptake rate).
 From the above, the difficulty level of driving is as follows.
Standard method: ① Low sludge concentration, ② Strict sludge management (pulling out) (narrow scope of proper sludge days), ③ Strict load (high load)/aeration balance (DO control), ④ Sludge condition prone to instability, ⑤ Narrow installation area, ⑥ Advanced management technology ⇨ Large urban sewage treatment plant.
Long-time method: ① High sludge concentration, ② Easy sludge management (withdrawal) (wide range of proper sludge days), ③ Easy balance of load (low load)/aeration ④ Stable sludge condition ⑤ Large installation area ⑥ No need for high management technology ⇨ Small sewage treatment plant, medium/large septic tank (including community drainage)
 From the above, when a beginner conducts an indoor experiment, it will be necessary to first master the long-term method.
Management of sludge days SRT
 In a laboratory experiment, a small capacity aeration tank will be used, so the sludge age control is performed by drawing the mixed solution from the aeration tank every day. Do not extract the returned sludge from the settling tank (since the sludge concentration is not constant).
 If the effective volume is 10 L, the SRT will be 10 days if 1 L of excess sludge is withdrawn from the mixture under aeration and stirring. If it is 0.5 L, it will take 20 days. If sampling is carried out every n days, SRT = 10/nX and nX [L] excess sludge will be extracted. The withdrawal interval can be daily or several days.
 The SRT may be 2 to 5 days for the standard method, 15 to 30 days for the long-term method, and the MLSS may be in the range of 1,500 to 2,000 mg/L for the former and 3,000 to 6,000 mg/L for the latter. Sludge management is limited to extraction of the mixed liquid, MLSS is left to the natural value, and no special adjustment is required. Regarding the standard method, strict control of the load and the sludge is required.

(3)汚泥の性状測定

(a) 沈降特性
 毎日あるいは数日おきに、SV30 の測定を行い、汚泥の沈降性を観察する。さらに、SVI を求める。沈降特性および SV30、SVI が適正な状況でない場合(SVI > 200)には、何か障害が生じている。
 熟練してくると、SV30 の値だけで、下記の MLSS 濃度が推定できるようになる。測定試料は 1.0 Lを要するので、実験混合液の容積が少ない場合(5 L以下)には、静置前に測定して、測定後、バケツに試料を戻す。試料採取中、バケツの曝気を行って均一なものを採取する。
(b) 汚泥濃度 MLSS の測定
 定期的( 1 週間に 1 回程度でよい)に汚泥濃度MLSSを測定する。減圧ろ過器・乾燥器が必要である。特に必要としない場合には、MLVSS の測定は不要である。目的の MLSS 値になるように、引抜量を増減して SRT を調整する。

(3) Sludge property measurement

(a) Settling characteristics
 Measure SV30 every day or every few days to observe sludge settling. Furthermore, the SVI is calculated. If the settling characteristics and SV30 and SVI are not in the proper condition (SVI > 200), something is wrong.
 As you become more skilled, you will be able to estimate the following MLSS concentrations using only the SV30 value. Since the measurement sample requires 1.0L, when the volume of the experimental mixture is small (5L or less), measure before the settling operation and return the sample to the bucket after measurement. During sampling, a bucket is aerated and a uniform sample is collected.
(b) Measurement of sludge concentration MLSS
 Measure the sludge concentration MLSS regularly (once a week is enough). A vacuum filter and a dryer are required. MLVSS measurement is not required unless otherwise required. Adjust the SRT by increasing or decreasing the withdrawal amount so that the desired MLSS value is obtained.

(4)曝気速度

 静置直前(1回サイクル/日の場合)に DO を測定し、DOが数mg/LであればOKである。時々、静置前の数時間前後の DO を測定し、図1に示すDOの上昇の有無を確認する。静置直前に、DO = 0 mg/Lであれば、曝気不足である。逆に、静置数時間以上も前に、DO が上昇していれば、過曝気となり、汚泥濃度が減少する。人間と同じで、食事と運動のバランスにより、太ったり・痩せたりする。

(4) Aeration rate

 DO is measured immediately before settling (1 cycle/day) and
it is OK if the DO is a few mg/L. Occasionally, measure DO for several hours before settling to check if DO rises as shown in Fig.1. If DO = 0mg/L immediately before settling, the aeration is insufficient. Conversely, if DO rises more than several hours before settling, it will be over-aerated and the sludge concentration will decrease. Like human beings, they gain or lose weight depending on the balance between diet and exercise.

曝気槽に BOD が高濃度のときは、BOD分解は酸素の供給速度が律速となり、DO 値は “0” である。BOD が消費され低濃度になると、NH4+の酸化が始まる。この条件では、(酸素供給速度) > (NH4+ 酸化速度) となり、少しずつ、DO が増加する。上記に記載の「DOの上昇」とは、この現象をいう。

When the concentration of BOD in the aeration tank is high, the rate of oxygen supply for BOD decomposition is rate-determining and the DO value is “0”. When BOD is consumed and the concentration becomes low, NH4+ oxidation starts. Under this condition, (oxygen supply rate) > (NH4+ oxidation rate), and DO increases little by little. The above-mentioned “DO rise” refers to this phenomenon

(5)pH の管理

 本サイトに記載の人工汚水(スキムミルク + 塩類)は、pH の調整機能があるので、pH 制御装置は不要である。しかし、pH が中性付近にあるか、時々、チェックする。もし、pH < 6 になるようであれば、NaHCO3(炭酸水素ナトリム、重曹)の割合を増量する。

(5) pH control

 The artificial wastewater (skimmed milk + salts) described on this site does not require a pH controller because it has a pH adjustment function. However, occasionally check that the pH is near neutral. If pH < 6, increase the ratio of NaHCO3 (sodium bicarbonate, baking soda).

(6)温度管理

 一般に哺乳動物は、体温調節ができる。その他の動物・植物は冬眠・夏眠したりして、それぞれの環境に対応した生活を送っている。
 細菌類や微生物は、環境温度によって「種」が変化する。また、1 日の温度差が 5°C を越えると生育が不安定となる。下水処理などでは、水量も多く、下水管・処理池は、地面下で保温条件もよく、1 日の温度差を少なく維持できている。
 ところが、室内実験では、特に冬期において、昼間(暖房-on)と夜間(暖房-off)の温度差が 20°C 以上になることも少なくない。この温度変化が、室内実験での活性汚泥を不安定にする主な原因である。屋外でも、冬期には温度差が大きい。
 冬期は固定温度ヒーター(25~30°C)をバケツ(曝気槽)内へ設置して、温度を一定にすることで、安定稼働が達成されると考える。このとき注意すべきは、水の蒸発、空だきである[ヒーターに自動電源切断(温度ヒューズ)の有無をメーカーに確認、火災防止のため)]。また、実験室の温度を昼夜連続管理していない場合には、下図に示す安価な恒温槽などに曝気槽を設置して、温度を一定してもよい。

(6) Temperature control

 Generally, mammals are capable of thermoregulation. Other animals and plants hibernate and fall asleep, and live their lives in accordance with their respective environments.
 The “species” of bacteria and microorganisms change depending on the ambient temperature. Moreover, growth becomes unstable when the temperature difference per day exceeds 5°C. In the case of sewage treatment, the amount of water is large, and the sewage pipes and treatment ponds are well maintained under the ground, and the temperature difference during the day can be kept small.
 However, in laboratory experiments, it is not uncommon for the temperature difference between daytime (room heating-on) and nighttime (room heating-off) to exceed 20°C, especially in winter. This temperature change is the main cause of destabilizing activated sludge in laboratory experiments. Even outdoors, the temperature difference is large in the winter.
 It is thought that stable operation will be achieved by installing a fixed temperature heater (25 to 30°C) in the bucket (aeration tank) in winter to keep the temperature constant. At this time, it is necessary to pay attention to water evaporation and emptying [check with the manufacturer whether the heater has automatic power off (thermal fuse), to prevent fire). Further, When the temperature in the laboratory is not continuously controlled day and night, an aeration tank may be installed in an inexpensive thermostatic chamber shown in the figure below to keep the temperature constant.

(7)曝気槽のサイズと形状

 以下、曝気槽容積とは混合液容量(有効容積)のことを示す。曝気槽の形状の要点は、①効率的な酸素移動、②十分な容器内の流動である。
 実施設では、一般的に、曝気槽の形状は矩形で、水深は 4m ~ 6m であり、散気管による曝気で酸素供給と混合液攪拌が行われる。家庭用小型浄化槽(構造例示形)の水深 1.2m 以上となっている。
 屋内実験の曝気槽の容積は、数十L以下で水深が浅く、散気による曝気では水中での滞留時間が短く、酸素移動効率が低く、かつ、気泡上昇による攪拌効率も悪い。
 以上のことから、曝気槽の形状については、下記の事項に留意する。

(7) Size and shape of aeration tank

 Hereinafter, the aeration tank volume refers to the mixed solution volume (effective volume). The main points of the shape of the aeration tank are ① efficient oxygen transfer and ② sufficient flow in the container.
 In an actual facility, the shape of the aeration tank is generally rectangular, and the water depth is 4m to 6m. Oxygen supply and mixture agitation are performed by aeration with a diffuser pipe. The water depth of a small household septic tank (exemplified structure in Japan) is 1.2 m or more.
 The volume of the aeration tank for indoor experiments is less than tens of liters and the water depth is shallow. The aeration by aeration has a short residence time in water, the oxygen transfer efficiency is low, and the agitation efficiency due to bubble rise is also poor.
 From the above, pay attention to the following matters regarding the shape of the aeration tank.

500mL~5L

 理化学用メスシリンダー(例えば、5L用:φ = 13cm, H = 60cm)など、円筒形で容量に応じて市販されいるものを利用するとよい。(半)透明であれば、プラスチック・ガラス製いずれでもよい。また、ペットボトルやブランデーボトルの上部を切り取って、曝気槽としてもよい。有効水深は容器高さの2/3程度が限度である。
○ 曝気容器:曝気槽の転倒防止のため、側面が格子状・底面が平底状の食器容器などを用い、園芸用ビニールタイ(巻線型)(同じ価格でも、品質に差があるので注意)で曝気槽容器のサイズに合わせて囲いを作り、これに収納する。多数の同時実験が可能となる。
○ 曝気(エアストーン):曝気は、酸素供給だけでなく、混合液の攪拌も兼ねているので、粗大気泡が望ましい。また、エアストーンが浮上しないように、比重のある棒などに固定して、容器の底に設置する。
○ 分岐管:曝気槽が小さいと、観賞魚用エアポンプでも流量が過大となることがあるので、分岐管(分岐コック外径とチューブ内径に留意)を用いて、複数の曝気槽へ空気を供給するとよい。また、逆に、下記に示す10L以上の曝気槽BOD負荷が高いときには、分岐管に複数のエアポンプを接続して送気量を上げてもよい。
 なお、曝気量の調節のため、分岐コックバルブの一つは、エア抜きとし、曝気槽へのエア供給量を調節する。
○留意事項:分岐管で複数の曝気槽へエア供給する場合には、曝気槽サイズおよび内容液の高さを同一にする。

500mL ~ 5L

 It is recommended to use a cylindrical physics and chemistry graduated cylinder (for example, for 5 L: φ = 13 cm, H = 60 cm), which is cylindrical and is commercially available depending on the volume. As long as it is (semi)transparent, either plastic or glass may be used. Further, the upper part of a PET bottle or a brandy bottle may be cut off to form an aeration tank. Make an enclosure according to the size of the aeration tank container and store it in this. Multiple simultaneous experiments are possible.
○ Aeration (air stone): Aeration not only supplies oxygen but also agitates the mixed liquid, so coarse bubbles are desirable. Also, to prevent air stones from rising, fix it on a rod with a specific gravity and install it on the bottom of the container.
○ Branch pipe: If the aeration tank is small, the flow rate may be too high even with an ornamental fish air pump, so use a branch pipe (take note of the branch cock outer diameter and tube inner diameter) to supply air to multiple aeration tanks. Good to do. On the contrary, when the BOD load is high in the aeration tank of 10 L or more described below, a plurality of air pumps may be connected to the branch pipe to increase the air supply amount.
 In order to adjust the amount of aeration, one of the branch cock valves is air bleed to adjust the amount of air supplied to the aeration tank.
○ Note: When supplying air to multiple aeration tanks using a branch pipe, make the aeration tank size and height of the content liquid the same.

10~20L前後

 本サイト内の別ページに示す写真1&2に示すような透明 PVC 製円筒型で、高さ H > 直径 φ であるような容器がよい。底面から適当な間隔で、バルブ付きコックを溶接すると、有効容積の可変できる。底面にはテーパ状として、排水バルブ付きコックを付けるとよい。

Around 10~20L

 A cylindrical container made of transparent PVC, as shown in Photos 1 and 2 on another page of this site, with a height H > diameter φ is preferable. The effective volume can be varied by welding the valve cock at an appropriate distance from the bottom surface. It is advisable to attach a drain valve cock to the bottom surface as a taper shape.

20L以上

 円形型は制作上コストが高くなるので矩形型となるが、なるべく、高さ H > 底辺 (W + D)/2 となるものがよい。背の高いバケツでもよい。
○攪拌:容器サイズが大きくなると、曝気泡による流動・攪拌が十分でないので、大きな羽根を低速回転(高価)で攪拌するとよい。この際に、センサーコードや散気管チューブなどが攪拌羽根に巻き込まないように、これらを細い塩ビパイプに通し、その塩ビ管を曝気槽側壁に固定するとよい(参照:回分法ページ図3)。
 また、図4に示す観賞魚用循環ポンプ(安価)を用いて攪拌してもよい。この場合には、曝気管(球)の反対側から吸込み、曝気管へ向けて吐出すように配置する。また、底面設置型の水中循環ポンプは、適切に槽内攪拌が行われる位置に据え付ける。これらのポンプの回転羽根に生物膜が付着するので、小まめに取り外して洗浄する。羽根の回転速度が速く、少しのブロック解体が起こるのが欠点であるが、沈殿槽でフロックの再集合が起こるので、汚泥分離に大きな障害を生じることはない。

20L or more

 Since the circular type is expensive in production, the rectangular type is used instead, but it is preferable that the height H > bottom (W + D)/2. A tall bucket is fine.
○ Stirring: When the size of the container becomes large, the flow and stirring due to the aeration bubbles are not sufficient, so it is advisable to stir the large blades at low speed (expensive). At this time, in order to prevent the sensor cords, diffuser tubes, etc. from getting caught in the stirring blades, pass them through a thin PVC pipe and fix the PVC pipe to the aeration tank side wall (see Fig.3 on the batch method page).
 Alternatively, the circulation pump for ornamental fish (inexpensive) shown in Fig.4 may be used for stirring. In this case, it is arranged so that it is sucked from the opposite side of the aeration pipe (ball) and discharged toward the aeration pipe. The bottom-mounted submersible circulation pump shall be installed at a position where agitation in the tank is performed appropriately. Since the biofilm adheres to the rotary vanes of these pumps, they are frequently removed and washed. The disadvantage is that the rotation speed of the blades is high and some block disassembly occurs, but since flocs are reassembled in the settling tank, there is no major obstacle to sludge separation.

留意事項

 いずれのサイズの曝気槽を用いる場合にも、望ましくは円筒形がよい。ホームセンターなど入手する場合で、ダスト容器などを用いる場合には、角が丸まったものがよい。室内実験での曝気槽では、嫌気ゾーンのない完全混合型が理想であり、混合液の適正な流動が求められる。市販品曝気槽(室内実験用)の中には、曝気用の隔壁や沈殿槽へ送液のための隔壁・越流堰などが設置(実施設をそのまま小型化)されているものもあるが、混合液の流動状態が悪く、様々なトラブルの原因となる。また、定期的な清掃作業が必要となるので、隔壁や付属品は邪魔でしかない

Notes

 When using an aeration tank of any size, a cylindrical shape is preferable. When using a dust container or the like obtained at a home improvement center, the one with rounded corners is preferable. In the aeration tank in the laboratory experiment, a perfect mixing type without an anaerobic zone is ideal, and proper flow of the mixed solution is required. Some commercially available aeration tanks (for laboratory experiments) have partition walls for aeration, partition walls for sending liquid to the settling tank, overflow weirs, etc. resulting that the fluid state of the mixed solution is bad and various troubles causes. In addition, since it requires regular cleaning work, the partition walls and accessories are only an obstacle.

4.活性汚泥の性状・機能の悪化と対策

 回分・連続に関わらず、汚泥の性状悪化には、大きく分けて、以下の 7 項目が挙げられる。内容を理解して、性状悪化に対しては実験方法や運転方法の見直しをする。

4. Deterioration of properties and functions of activated sludge and countermeasures

 Regardless of whether it is batch or continuous, deterioration of sludge properties can be broadly divided into the following 7 items. Understand the contents and review the experimental method and driving method for deterioration of property.

(1)泥水などに含まれるコロイド状無機微粒子の不足

 下水などには、洗濯水、床洗浄水、排水溝、雨水などが含まれ異物・砂・泥などの成分が含まれる。異物や砂などは、スクリーン・沈砂池・初沈池で除去されるが、無機微粒子・髪・繊維ごみ(以下、微粒子等と省略)などは曝気槽へ流入する。活性汚泥と微粒子等が堅牢な混合フロックを形成し、さらにフロック比重が増加するので、沈降性のよい良質な活性汚泥となる。
 シリンダーで沈降状況を観察すると、上澄み液が透明な界面沈降で沈降も速く、5~10 分程度で圧縮沈降へ移行する。(参考ページ:沈殿池の基礎)。
 天候・季節によって変動するが、下水汚泥には、20~30% の無機物質(強熱残量)が含まれている。下水汚泥の焼却灰は厄介者であるが、下水処理においては、活性汚泥の良好な沈降分離に役立っている。しかしながら、下水中の汚泥中有機物の割合は、年々増加傾向にあり、有機物割合が90%弱の処理場も見受けられるようになった。これは食生活の変化、即ち脂質や動物性タンパク質の摂取量の増加、および下水排除方式が分流式になってきた。即ち下水に流入する無機質が減少してきた事等によると思われる。[川崎、1985]
 室内実験などで、模擬汚水を投与する場合には、上記の微粒子等が含まれていないので、下水汚泥を「種」として実験すると、7~20 日程度で上記の微粒子等が系外に排出され、少しずつ汚泥フロックが軽くなり自由沈降の状態となって、上澄み液も懸濁し、沈降速度も遅くなる。

(1) Lack of colloidal inorganic fine particles contained in muddy water, etc.

The activated sludge and the fine particles form a robust mixed floc, and the specific gravity of the floc increases, so that the activated sludge of good quality has good settling properties.
 Observing the settling situation with a cylinder, the supernatant liquid is a transparent with clear interface and the settling is fast, and it shifts to compression settling in about 5 to 10 minutes. (Reference page: Basics of Settling Tank/Basin).
 Depending on the weather and season, sewage sludge contains 20 to 30% of inorganic substances (residue after ignition). Incineration ash of sewage sludge is a nuisance, but in the treatment of sewage, it contributes to good settling and separation of activated sludge. However, the proportion of organic matter in sludge in sewage has been increasing year by year, and some treatment plants have an organic matter proportion of less than 90%. This is due to changes in dietary habits, that is, increased intake of lipids and animal proteins, and sewage elimination systems have become divergent. That is, it is considered that the amount of minerals flowing into the sewage has decreased. [Kawasaki, 1985]
 When artificial sewage is feeded in laboratory experiments, etc., the above fine particles are not included. Therefore, if sewage sludge is used as a “seed”, the above fine particles will be discharged from the system in about 7 to 20 days. The sludge flocs become lighter little by little and become a free settling state, the supernatant liquid also suspends wuthout clear interface, and the settling speed slows down.

カオリン投与とその添加量の計算事例

 下水などの実汚水でなく、模擬汚水、あるいは、無機微粒子等を含まない汚水を用いる場合には、活性汚泥中の微粒子等を常に 20% 程度に維持する必要がある。安価な化粧用カオリンなどを利用するとよい。
 曝気槽内の汚泥・総重量の 20% 程度に相当するカオリンを定期的に投入する。基本的な原理は、汚泥日令が経過すると、全てのカオリンが消失するものとする。本サーバにカオリン投与量の計算xlsファイルがあるので、ダウンロードして活用するとよい。
○ 曝気槽の有効容積 V: 10L
○ 汚泥日令 SRT: 20日
 長時間曝気法: SRT = 曝気槽内の混合液を0.5L/d抜取 = 10L/(0.5L/d)=20d
 休祭日がn日のときには、その前後日に0.5L/d✕(n+1)抜き取る
 汚泥濃度 MLSS: 3,000mg/L(直近の5回程度の測定値の平均値)
○ 7日の間隔(曜日を決めておく)

○ 10日の間隔(10日、20日、30日のように投与する日を決めておく、休祭日には、その前後日とする)

Calculation example of kaolin administration and its addition amount

 When using simulated sewage or sewage that does not contain inorganic fine particles, etc., instead of actual wastewater such as sewage, it is necessary to always maintain the fine particles in activated sludge at around 20%. Use cheap cosmetic kaolin.
 Sludge in the aeration tank, kaolin equivalent to about 20% of the total weight is periodically added. The basic principle is that all the kaolin will disappear when the sludge age has passed. This server has a calculation xls file for kaolin dosage, which you can download and use.
○ Effective volume of aeration tank V: 10L
○ Sludge day SRT: 20 days
 Long aeration method: SRT = 0.5L/d withdrawal of mixed liquid in aeration tank = 10L/(0.5L/d) = 20d.
 When there are n holidays, remove 0.5L/d✕(n+1) days before and after that.
 Sludge concentration MLSS: 3,000mg/L (average of the most recent 5 measurements).
○ Interval of 7 days (determine the day of the week.).
 3,000[mg/L]✕10[L]/20[d]✕7[d]✕20[%] = 2.1g.
○ Interval of 10 days (determine the administration day such as 10th, 20th, 30th, and on holidays, use the days before and after that.).

Simulated wastewater (skimmed milk + nutrient salts + pH adjuster): 300mg-BOD/L, HRT = 24h, MLSS = 1,800mg, MLSS at SV30 = 7,200mg/L (at the start of the experiment), SRT = 20d.
VSS/SS is constant at 90% after 20 days. The ignition residue after 10 days is 10%, but it is a soluble inorganic salt in simulated sewage, not a component derived from colloidal fine particles.

(2)油分を含む排水

 油分を含む汚水を処理すると活性汚泥の沈降性が悪くなる。多数ある料理店街など高濃度油分を含む下水や油分濃度の高い食品工場排水の処理では、凝集助剤(粘土系、アルミ・鉄系など)・高分子凝集剤を投入し、処理コストが高くなる。排出源におけるオイルトラップなど設置とその適正な維持管理が必須となる。さらに、加圧・浮上分離による油分の除去を行う。

(2) Wastewater containing oil

 When the wastewater containing oil is treated, the settling property of the activated sludge becomes worse. When treating sewage containing high-concentration oil and food factory wastewater with high oil content, such as in many restaurants, the inorganic coagulant (clay-based, aluminum/iron-based, etc.) and polymer coagulant are added, resulting in high treatment cost. It is essential to install an oil trap at the emission source and maintain it properly. Furthermore, oil content is removed by pressurization and floating separation.

(3)糸状細菌の発生

 季節の変わり目、高温・低温時期、活性汚泥の弱体化(過曝気・酸素不足、負荷の過不足、栄養塩類の過不足、阻害物質や油分の混入など)など、様々な原因により発生する。
 糸状菌の発生原因を解明し、根気よく改善に努めることも経験となる。一方で、実験室では最初からやり直したほうが効果的な場合もある。

(3) Occurrence of filamentous bacteria

 It occurs due to various causes such as seasonal changes, high or low temperature periods, weakening of activated sludge (excessive or insufficient aeration, excess or insufficient load, insufficient nutrient salts, mixing of inhibitors and oils, etc.).
 It is also an experience to elucidate the cause of filamentous bacteria and make patient efforts to improve them. On the other hand, in the laboratory, it may be more effective to start over from the beginning.

Since the outer wall of the settling tank is dirty, it is difficult to see the state of the supernatant, but it is extremely transparent and clean. However, sludge swells and sedimentation becomes extremely difficult. It is necessary to reduce the load (especially the flow rate) and take a long-term response before recovering. If the aeration tank temperature is operated at room temperature, it tends to occur during the seasonal change period from autumn to winter.

This activated sludge has an SV30 of 98 to 100, and the MLSS of the aeration tank and the SS of the separated liquid in the settling tank are almost the same.

(4)過曝気(痩せる)・酸素不足(肥満)による汚泥の活性低下とフロックの解体

(a) 連続曝気

 新たに模擬汚水を投与( 1 日 1 回)する時刻前の 1~2 時間前後に、DO の上昇が起こる曝気速度とする。曝気の過不足には注意する。
 また、活性汚泥の弱体化、または、負荷(模擬下水成分の濃度)の過不足は、酸素消費量の変化(次サイクルの 1 時間前の DO 変化の有無)によって判断できる。曝気量の調節のみでなく、上記の DO 状況によって、負荷を増加/減少することも大切である。
フロックの弱体化
 バクテリアの細胞表面は、-COOH ⇄ COO 基によって、pH > 4 ではマイナス電荷を帯びている。(なお、アミノ酸のアミノ基はプラス電荷を帯びているが、内部にあって他の構成物質、例えば、-OH などと水素結合している。詳しくは、本サーバー内のxlsファイルをダウンロードして、アミノ酸のシートを参照されたい。)。これによってバクテリアは、水中において分散状態で (下記ページを参照)、生存している。ところが、活性汚泥のバクテリア(凝集性)は細胞外に粘質物質を分泌して、互い付着・凝集してフロックを形成し、沈降性を有しています。流れのある地表水・海水中に生息する付着性バクテリアも同様である。
 自然界では、沈降すると酸素や栄養分を摂取できなくなるので、粘質物質を分泌しない分散性バクテリアが優占種となる。
 一方、活性汚泥法では曝気・機械攪拌により、フロックが水中で分散状態となるので、凝集性バクテリアの生存が可能となる。分散性バクテリアは沈降性がないので、最終沈殿池において放流水とともに系外へ排出される。凝集性バクテリアは沈殿分離して返送され、曝気槽に保持される。このように活性汚泥法とは、粘質物質を分泌するバクテリアを選択的に保持培養するシステムで、一般の分散性バクテリアは生存できない。
 フロックの弱体化(活性汚泥の解体)とは、不適切な環境におかれた凝集性バクテリアがその活性度の低下によって粘質物の細胞外分泌量が減少し、活性汚泥バクテリアの相互付着・凝集力が低下して分散状態となる現象をいう。

(4) Decrease in sludge activity and dismantling of flocs due to excessive aeration (thinning) or insufficient oxygen (obesity)

(a) Continuous aeration

 The aeration rate is set at which DO rises about 1 to 2 hours before the time of newly cycle (once a day) starts. Be careful of excess and deficiency of aeration.
 In addition, weakening of activated sludge or excess or deficiency of load (concentration of simulated sewage component) can be judged by changes in oxygen consumption (whether DO changes 1 hour before the next cycle). It is important not only to control the amount of aeration but also to increase or decrease the load according to the DO situation described above.
Flock weakening
 The cell surface of bacteria is negatively charged at pH > 4 due to the -COOH ⇄ COO group. (Although the amino group of an amino acid has a positive charge, it has an internal hydrogen bond with other constituents, such as -OH. For more information, download the xls file on this server., see the sheet of amino acids). This allows the bacteria to survive in a dispersed state in water (see page below). However, the activated sludge bacteria (coagulability) secrete mucous substances to the outside of the cell, attach and aggregate with each other to form flocs, and have sedimentability. The same applies to adherent bacteria that live in flowing surface water and seawater.
 In the natural world, when settling, oxygen and nutrients cannot be taken in, so dispersive bacteria that do not secrete mucilages become the dominant species.
 On the other hand, in the activated sludge method, flocs are dispersed in water by aeration and mechanical agitation, so that flocculating bacteria can survive. Since dispersible bacteria have no settling property, they are discharged out of the system along with the discharged water in the final settling basin. Aggregating bacteria are separated by sedimentation and returned to the aeration tank. As described above, the activated sludge method is a system for selectively retaining and culturing bacteria that secrete mucous substances, and general dispersible bacteria cannot survive.
 Weakness of flocs (disassembly of activated sludge) is the decrease in the activity of cohesive bacteria placed in an inappropriate environment, resulting in a decrease in the extracellular secretion amount of mucilage. Is a phenomenon in which the dispersion is lowered and the dispersion state is achieved.

(b) DO 計を用いた曝気量制御の場合
 2 台のエアポンプを使用し、1 台はやや不足の曝気速度、1 台は On-Off 制御する。DO 計は、定期的に校正し、劣化した電極(消耗品である)は更新する。また、酸化還元剤によって、異常な反応をするので、注意する。設定値は 数mg-DO/L とし、正確に設定しなくてよい。DOを 0 mg 近くの値に設定すると、DO 計の校正不良のとき、酸素不足となることがある。

(b) In case of aeration control using DO meter
 Two air pumps are used, one with slightly insufficient aeration rate and one with on-off control. DO meter should be calibrated periodically and the deteriorated electrode (consumable item) should be updated. Also note that the redox agent causes an abnormal reaction. The setting value is a few mg-DO/L, and it is not necessary to set it accurately. If the DO is set to a value near 0 mg, oxygen may be insufficient when the DO meter is calibrated incorrectly.

(5)発泡現象

 活性汚泥の生育環境(温度、投与する原水組成と負荷、新たな植種汚泥など)、少しずつの変化では、問題とならないが、これらの生育環境が急激に変化すると、必ず、発泡が生じる。市販品の曝気槽では、水面上部の側壁の高さが 5cm 程度のものもある。この高さでは、僅かな発泡でも、汚泥が流出する。写真1および写真2に示すように、少なくとも、曝気槽の水面から30cm以上の側壁が必要である。ただし、この発泡現象は、異常な汚泥の性状悪化ではなく、活性汚泥の生物相がその環境に順応できるようになると、発泡は減少してくる。
 放線菌が異常発生すると、頑固な発泡現象が生じる。放線菌の表面はミコール酸で覆われており、これによって菌体を保護している。ミコール酸は、炭素数60~90から構成される超高級脂肪酸である。したがって、これが異常に増殖すると、菌体から剥離したミコール酸が水中に分散して、界面活性剤(炭素数10~20程度)のような「サラサラ」した泡ではなく、粘性の高い頑固な泡となる。下記 5.1-(2)-5)発泡対策 に記載する消泡剤を多量に散布すると、一時消泡するが、多量に加えた消泡剤とミコール酸の相互作用により、粘性の高い混合液となり、返送汚泥ポンプの閉塞など、様々な障害が連鎖的に生じる。

(5) Foaming phenomenon

 Small changes in the growth environment of activated sludge (temperature, composition and load of raw water to be administered, new planted sludge, etc.) do not pose a problem, but abrupt changes in these growth environments always cause foaming. In some commercially available aeration tanks, the height of the side wall above the water surface is about 5 cm. At this height, even a slight foaming will cause sludge to flow out. As shown in Photo 1 and Photo 2, at least a side wall of 30 cm or more from the water surface of the aeration tank is required. However, this foaming phenomenon is not an abnormal deterioration of sludge properties, but foaming decreases when the biota of activated sludge becomes able to adapt to its environment.
 When actinomycetes occur abnormally, a stubborn foaming phenomenon occurs. The surface of actinomycetes is covered with mycolic acid, which protects the cells. Mycolic acid is an ultra-higher fatty acid composed of 60 to 90 carbon atoms. Therefore, if it grows abnormally, the mycolic acid exfoliated from the cells disperses in the water, and it is not a “smooth” foam like a surfactant (about 10 to 20 carbon atoms), but a highly viscous and stubborn foam. Becomes When a large amount of the defoaming agent described in 5.1-(2)-5) Foaming countermeasures below is sprayed, the foam defoams temporarily, but due to the interaction between the large amount of defoaming agent and mycolic acid, it becomes a highly viscous mixture. Various obstacles such as blockage of return sludge pump occur in a chain.

(6)汚泥浮上

 維持管理が良好な条件(曝気量が十分なとき)では、アンモニアが硝酸に酸化される。活性汚泥には、窒素還元菌(通性嫌気性細菌)が混在しているので、沈殿槽での汚泥の滞留時間が長くなると、窒素ガスが発生し、その浮力により汚泥が浮上する。この様な汚泥浮上は、汚泥の性状悪化ではない。沈殿池の汚泥全体を 1 rpm程度の速度で攪拌するとともに、返送速度を上げることにより、汚泥浮上を避けることができる。
実験に用いる供試汚泥
 下水処理施設から汚泥を入手して、沈降特性や濃縮薬剤効果を試験するときの留意すべきは事項は、試験中における汚泥の分離浮上である。例えば、汚泥特性を測定中、30分も経過すると汚泥が浮上して測定が困難となる場合が少なくない。これは、次の理由による。
 以前においては、下水処理はBOD除去を主目的に稼働していたが、閉鎖性水域の富栄養化などを理由に、窒素規制が厳しくなり、硝化・脱窒が行われるようになり、無酸素-好気(窒素除去)、嫌気-好気(リン除去)、嫌気-無酸素-好気(窒素・リン除去)など、汚泥の滞留時間が長くなって、硝化菌が十分増殖する環境となっている。このため、汚泥沈降測定中に無酸素状態となると脱窒反応が直ちに進行して、汚泥細胞内に窒素ガスが生成して汚泥が浮上する。
 このような活性汚泥に対しては、曝気槽の混合液の1/3程度を1週間~10日(温度 25℃、SRT 3サイクル程度)、毎日、抜き取るとよい。硝化菌が洗い流されて、汚泥浮上が起こらなくなる。
 また、下記の6(3)に記載しているように供試汚泥の洗浄を行うとよい。

(6) Sludge surfacing

 Under conditions of good maintenance (when the amount of aeration is sufficient), ammonia is oxidized to nitric acid. Since nitrogen-reducing bacteria (facultative anaerobic bacteria) are mixed in the activated sludge, if the sludge’s residence time in the settling tank becomes long, nitrogen gas is generated and the sludge floats due to its buoyancy. Such sludge flotation does not deteriorate sludge properties. The sludge floating can be avoided by stirring the whole sludge in the sedimentation tank at a speed of about 1 rpm and increasing the return speed.
Sample sludge used in the experiment
 When obtaining sludge from a sewage treatment plant and testing the sedimentation characteristics and the effect of concentrated chemicals, a matter to be noted is the separation and flotation of sludge during the test. For example, during the measurement of sludge characteristics, it often happens that the sludge floats up after 30 minutes and the measurement becomes difficult. This is for the following reason.
 In the past, sewage treatment was mainly operated to remove BOD, but due to factors such as eutrophication in closed water areas, nitrogen regulations became stricter and nitrification and denitrification began to take place, resulting in the long sludge retension time such as aerobic-anoxic conditions (nitrogen removal), anaerobic-aerobic conditions (phosphorus removal), anaerobic-anoxic-aerobic conditions(nitrogen/phosphorus removal). Therefore, if anoxic state occurs during sludge sedimentation measurement, the denitrification reaction immediately proceeds, nitrogen gas is generated in the sludge cells, and the sludge floats.
 For such activated sludge, about 1/3 of the mixed liquid in the aeration tank should be withdrawn daily for 1 week to 10 days (temperature 25°C, SRT 3 cycles). Nitrifying bacteria are washed away, and sludge floating does not occur.
 Moreover, it is advisable to wash the sample sludge as described in 6(3) below.

(7)ミミズの異常発生

 自然界では、植物プランクトンを動物プランクトンが補食し、その動物プランクトンを二次消費者が餌としている。有機物の分解者である細菌をアメーバやゾウリ虫、ツリガネ虫などの原生動物が捕食し、それらをワムシ類やミミズなどの後生動物が食べるという一連の食物連鎖が生じている。水域では魚類の餌として、陸地では肥沃な森林・農地の適正な土壌形成に極めて重要な役割を担っている。また、この捕食作用を積極的に余剰汚泥量を削減する汚濁水の浄化法もある。
 室内実験においても、ミミズの発生そのものは、水質が良好な指標であるが、時には異常発生して、活性汚泥が極端に減少することがある。対策としては、混合液をガーゼや台所細目ネットでの捕捉・駆除する。

(7) Abnormalities of earthworms

 In nature, phytoplankton are supplemented by zooplankton, and the zooplankton are fed by secondary consumers. Protozoa such as amoeba, elephants, and vines prey on bacteria, which decompose organic matter, and metazoans such as rotifers and earthworms eat them. As a bait for fish in water, it plays an extremely important role in proper soil formation in fertile forests and farmlands on land. There is also a method of purifying polluted water that positively reduces the amount of excess sludge by this predation action.
 In laboratory tests, the water quality is a good indicator of earthworm generation, but sometimes abnormalities occur and activated sludge extremely decreases. As a countermeasure, capture and exterminate the mixed solution with gauze or kitchen fine net.

5.糸状細菌対策ー短縮SRT法/粉末添加法/次亜塩素酸ナトリウム法

5.1 短縮SRT法

 簡単な方法として、SRTの短縮である。上記に示す標準活性汚泥のBOD負荷とし、SRT 3~4日程度で運転しSRTを数サイクル経過すると、糸状細菌は洗い流されて活性汚泥は正常な状態となることが多い。

5. Measures against filamentous bacteria – shortened SRT method/powder addition method/sodium hypochlorite method

5.1 Shortened SRT method

 An easy way is to shorten the SRT. When the standard activated sludge shown above is loaded with BOD and the SRT is operated for about 3 to 4 days and several SRT cycles have passed, the filamentous bacteria are often washed away and the activated sludge is in a normal state.

5.2 スキムミルク混合粉末添加法

(1)適用範囲

 本粉末法は、あくまでも、屋内実験にのみ適用できるもので、実処理施設に対しては不可能である。
 (a) 植種用汚泥に異常に糸状細菌が含まれていたり、上記の4(3)に示した原因等で糸状細菌が発生し、汚泥膨化・発泡などにより実験の継続が困難になったときに適用する。
 (b) 現場の実廃水や他組成の人工廃水を実験中で、糸状細菌の発生した実験汚泥を回復させたい場合にも、適用できる。
 (c) また、実処理施設での活性汚泥の性状が悪化したときの原因解明の一つとしても、有効な検証法である。
 本法を適用し、糸状細菌が消滅してから、個々の実廃水や人工廃水へ切り換える。元の実験に復帰して、再度、糸状細菌が発生する場合には、実験系に不具合があるので、その原因を検討すべきであろう。
 本法の特徴は、汚泥流出がないので、初心者は安心して実験ができる。唯一のトラブルは、下記の5)発泡対策のみである。注意すべき事項は、曝気速度(1mg<DO<3mg)と容積負荷(g-BOD/L/d)のバランスのみである。過曝気では汚泥の解体が進行し、酸素不足では汚泥の膨化が起こる。長時間(半日以上)に及ぶ酸素不足では汚泥腐敗やカビ発生の原因となることがある。また、適正なDO管理が行われても、曝気槽内に滞留部分があると、長時間の過曝気と酸素不足のゾーンが生じるので、上記の「曝気槽のサイズと形状」で示した容器内の流動状態には、特に注意する。

5.2 Skim milk mixed powder addition method

(1) Scope of application

 This powder method can be applied only to indoor experiments, not to actual treatment facilities.
 (a) Abnormal filamentous bacteria are contained in the sludge for planting, or filamentous bacteria are generated due to the reasons described in “4(3)” above, causing sludge swelling and foaming. It is applied when it becomes difficult to continue the experiment.
 (b) It can also be applied when actual wastewater at the site or artificial wastewater of other composition is being tested and it is desired to recover the experimental sludge in which filamentous bacteria have been generated.
 (c) It is also an effective verification method as one of the reasons for clarifying the cause of deterioration in the properties of activated sludge in an actual treatment facility.
 By applying this method, after the filamentous bacteria have disappeared, individual wastewater or artificial wastewater is switched to. If filamentous bacteria are generated again after returning to the original experiment, the experimental system has a problem, and the cause should be examined.
 The feature of this method is that there is no sludge outflow, so beginners can experiment with peace of mind. The only trouble is the following “5) Measures against foaming“. The only thing to be careful about is the balance between aeration rate (1mg < DO < 3mg) and volumetric load (g-BOD/L/d). With excessive aeration, sludge dismantling proceeds, and with insufficient oxygen, sludge swelling occurs. Lack of oxygen for a long time (more than half a day) may cause sludge decay and mold formation. Even if proper DO management is performed, when there is a stagnation in the aeration tank, over-aeration and oxygen-deficient zones will occur for a long-time , so pay particular attention to the fluid state inside the container as shown "3.(7) Aeration tank size and shape” above.

(2)実施法

1)スキムミルク混合粉末の調整
 サーバーに保存中のxlsファイルダウンロードし、これに(以下、粉末と略称)準じて、スキムミルク・栄養塩類・pH調整剤を混合して、所定量の混合粉末を調整し、保管する。
2)回分法
 連続活性汚泥法では、原水投入を停止して、回分法に切り換える。なお、汚泥返送ポンプは稼働し、沈殿槽での HRT を 1~2h 程度になるように、返送循環する。
3)DO制御と曝気量の調整
 DO制御装置が付設されている場合には、1mg/L<DO<3mg/L に設定する。DOの上限は3mg/Lに近い値であれば、問題ない。DO制御器がない場合には、図1に示したように曝気量を調整する。
4)粉末投与
 曝気槽・容積、SRT、MLSS、曝気速度等から、適宜、粉末投与量を決定する。図1に示した手順に沿って、回分法を運転する。
5)発泡対策
 粉末投与直後に発泡するので、下記のように措置する。
 溶液型シリコン系消泡剤の 5 倍希釈液をスプレー(JET↔SPRAYの調整可能で、200~500mL程度、100円ショップ等で販売)のJET状で、曝気槽表面に散布する。数mL の散布で消泡できる。
6)測定・観察項目と維持管理
 次の項目の測定を行い、粉末投与量または曝気速素を図1の状況になるように調整する。
 ① DO:朝、昼、夕に3回/日
 ② SV30・MLSS:1回/日
注意)連続曝気法を粉末回分法へ変更した場合には、沈殿槽の界面が1/2~1/3(ただし、糸状菌が減少し回復過程にあるとき)になるように汚泥返送速度を調節する。毎日、沈殿槽の汚泥界面の高さ、界面の明確度、上澄液の透視度など観察し、汚泥回復の状況を調べる。
 ③ 温度図4に示す熱帯魚水槽のヒーター(100W )を曝気槽へ浸漬して、温度26℃程度に保つ。
 ④ 同粉末は、pH緩衝能力が高く、海水pHと同程度に保たれる。念のため、適宜、pHも測定するとよい。
 ⑤ 粉末投与量と曝気量:DO計測制御器を用いて曝気量を制御して運転する場合には、粉末投与量は適宜、実験条件に応じて選定できる。同機器がなくて一定曝気量で運転する場合には、図1に示すDO挙動を示すように粉末投与量に対応した曝気量を設定する必要があり、この調節に手間がかかる。
 ⑥ MLSS:本粉末法では、汚泥の流出がないので、最大 MLSS 8,000mg/L 程度まで運転可能であるが、6,000mg/Lを越えるときには、汚泥抜取量を増加しSRTを短縮するとよい。
7)糸状細菌の消滅
 粉末投与・回分法により適正に運転した条件化では、異常に発生した糸状細菌が消滅するまで20~30日が必要である。可能であれば、顕微鏡で糸状細菌の状態を観察するとよい。50<SVI<150が望ましいが、SVI<200 または SV30<60~80、かつ、汚泥界面が明確になれば、糸状細菌はほぼ消滅し、回復したと判断する。


スキムミルク混合粉末の特徴
スキムミルク(脱脂粉乳)は、炭水化物とタンパク質のバランスがよく、脂質が少なく、ビタミン・ミネラルも豊富である。また、BOD:N:Pの比率も適正である。NaHCO3・NaCO3が多量に含まれているので、pH緩衝力が強く、曝気液のpHは、表面海水のpH(8.1~8.2)付近に維持される。BOD[g] ≒ (1/2)×スキムミルク [g] で、BOD負荷の計算が簡単である。以上のことから、初心者が活性汚泥実験を行うときの、人工廃水として適している。
(2) Implementation method

1) Preparation of skim milk mixed powder
Download the xls file stored in the server, mix skim milk, nutrient salts, and pH adjuster according to this file to prepare a predetermined amount of mixed powder and store it (hereinafter, abbreviated as powder).
2) Batch method
 In the continuous activated sludge method, the raw water input is stopped and the batch method is separately used. In addition, the sludge return pump operates and the return circulation is performed so that the HRT in the settling tank is about 1 to 2 hours.
3) DO control and adjustment of aeration amount
 If a DO controller is installed, set 1mg/L < DO <3mg/L. There is no problem if the upper limit of DO is a value close to 3mg/L. If there is no DO controller, adjust the aeration amount as shown in Fig.1.
4) Powder administration
 Determine the powder dose appropriately from the aeration tank/volume, SRT, MLSS, aeration rate, etc. The batch method is operated according to the procedure shown in Fig.1.
5) Foaming measures
 Since foaming occurs immediately after powder administration, take the following measures.
 Spray 5 times diluted solution type silicon type antifoaming agent on the surface of the aeration tank in the form of JET spray (JET ↔ SPRAY adjustable, about 200 to 500mL, sold at 100 yen shops etc.). It can be defoamed by spraying a few mL.
6) Measurement/observation items and maintenance
 Measure the following items and adjust the powder dose or the aeration quickener so that the situation shown in Fig.1 is obtained.
 ① DO: Morning, noon, and evening 3 times/day.
 ② SV30 and MLSS: 1 time/day.
Note) When the continuous aeration method is changed to the powder batch method, the sludge return rate should be set so that the interface of the settling tank becomes 1/2 to 1/3 (however, when the filamentous fungus decreases and is in the process of recovery). Adjust. Every day, observe the height of the sludge interface in the settling tank, the clarity of the interface, the transparency of the supernatant, etc. to check the status of sludge recovery.
 ③ Temperature: The heater (100W) of the tropical fish tank shown in Fig.4 is immersed in the aeration tank to keep the temperature at about 26°C.
 ④ The powder has a high pH buffering capacity and is maintained at the same level as seawater pH. As a precaution, pH should be measured appropriately.
 ⑤ Powder dose and aeration rate: When operating with the DO measurement controller controlling the aeration rate, the powder dose can be appropriately selected according to the experimental conditions. When operating with a constant aeration amount without the same device, it is necessary to set the aeration amount corresponding to the powder dose as shown in the DO behavior shown in Fig.1, and this adjustment takes time.
 ⑥ MLSS: With this powder method, there is no sludge outflow, so it is possible to operate up to a maximum of MLSS of 8,000mg/L. However, when it exceeds 6,000mg/L, it is advisable to increase the sludge withdrawal amount and shorten the SRT.
7) Disappearance of filamentous bacteria
 Under the condition of proper operation by the powder batch method, it takes 20 to 30 days until the abnormally generated filamentous bacteria disappears. If possible, it is advisable to observe the state of filamentous bacteria with a microscope. 50 < SVI < 150 is desirable, but if SV30 < 60 to 80 or SVI < 200 and the sludge interface becomes clear, it is judged that the filamentous bacteria have almost disappeared and the sludge is recovered.


Features of skim milk mixed powder
Skim milk has a good balance of carbohydrates and proteins, is low in fat, and is rich in vitamins and minerals. Also, the ratio of BOD:N:P is appropriate. Since it contains a large amount of NaHCO3 and NaCO3, it has a strong pH buffering power, and the pH of the aeration liquid is maintained near the surface seawater pH (8.1 to 8.2). BOD[g] ≈ (1/2) × skim milk [g], and the calculation of BOD load is easy. From the above, it is suitable as an artificial wastewater when a beginner conducts an activated sludge experiment.

5.3 次亜塩素酸ナトリウム法

執筆予定
 下水道の終末処理場の糸状細菌の発生対策については、横浜市のHP「下水道調査・研究 運転管理に関する調査・研究」に詳細が記載されているので、該当ページを参照されたい。
 屋内実験系への本法の適用にあたっては、基礎的な実験・測定技術が必要であるので、関連ページを掲載の後、執筆する。

6.生物膜法

(1)維持管理の簡素化

 生物膜法(生物ろ過法、浸漬ろ床法)法は、負荷変動・温度変化に対して、比較的に安定な稼働をする。実験目的によっては、採用すると便利である。例えば、難分解性汚水の処理や阻害物質の有無など。
 下記の文献 2)に示す装置は、1 日 1 回、または数日1回の排水・投入、定期的な水質検査でよい。負荷量は、ろ床の再生回数(1~2 週間に 1 回程度)に合わせて行う。曝気の必要はない。検討され、有効であれば活用されとよい。

6. Biofilm method

(1) Simplification of maintenance
 The biofilm method (biological filtration method, immersion filtration method) operates relatively stably against load fluctuations and temperature changes. Depending on the purpose of the experiment, it may be convenient to adopt. For example, treatment of persistent degradable sewage and the presence or absence of inhibitors.
 The equipment shown in reference 2) below, the water treated is drained and the raw wastewater is inputed once a day or once every several days, and regularly tested for water quality. The amount of load should be adjusted according to the number of times the filter bed is regenerated (about once every 1 to 2 weeks). No need for aeration. It should be examined and, if effective, utilized.

(2)多数の検証実験

 (a) 阻害物質などの影響を調べる場合には、濃度効果の実験数に応じて、台数を設置することとなる。
 阻害物質の影響(慢性)の結果取得まで、少なくとも、数ヶ月が必要である。急性影響は 1~数週間程度となる。設置場所、経費節減(装置・管理費)、台数多数の場合には、下記に示す外付けフィルター付き観賞魚用小型水槽を用いてもよい。数日~1 週間に 1 回程度のフィルター再生。フィルターを 2 台設置するとよい。
 (b) この実験で、阻害効果を確認したのち、このバケツ等による活性汚泥の回分実験を行う。このとき、「種」は、生物膜法(生物ろ過法)のろ床の汚泥を使用する。
 (c) 実験装置は、熱帯魚用小型水槽を活用するとよい。この場合には、適宜、ろ過装置のフィルターを洗浄再生する。水槽へ底砂などを敷かない場合には、ろ過装置を 2 台とする。フィルターを洗浄すると、バクテリアが存在しなくなるので。

(2) Many verification experiments

 (a) When investigating the effects of inhibitors, etc., the number will be set according to the number of experiments of concentration effect.
 At least several months are required to obtain the results of the effects of inhibitors (chronic). The acute effect is about one to several weeks. If there are many installation sites, cost savings (equipment/management costs), and a large number of units, the small aquarium for ornamental fish with an external filter shown below may be used.  (b) After confirming the inhibitory effect in this experiment, a batch experiment of activated sludge using this bucket or the like will be performed. At this time, the “seed” uses sludge on the filter bed of the biofilm method (biological filtration method).
 (c) The experimental device should utilize a small tropical fish tank. In this case, the filter of the filtration device is appropriately washed and regenerated. If the bottom tank is not covered with sand, use two filtration devices. Bacteria are gone when you wash the filter.

(3)実験用汚泥の育成

 観賞魚用60L水槽セット(但し、浄化フィルター:2台、フィルター充填剤:大磯砂 3~5mm 前後、底砂:左記の大磯砂を厚さ 3~5cm 敷き詰める)を利用するとよい。上記スキムミルク粉末を15~20g/日を投与すると、浄化フィルターの再生は、交互に、5日程度の間隔で、良質な供試汚泥 1L程度(MLSS 8,000~10,000mg/L、SVI 50mL/g 前後)が得られる。
 適正な粉末投与量は、翌日の粉末投与時において、水質が透明であればよい。懸濁している場合には、投与量を削減する。水槽の水交換は、毎週1回程度、水槽水の1/2を水道水で交換する。スキムミルク投与の場合には、pH 調整は不要である。
 供試汚泥の採取は、浄化フィルターがオバーフローしたら、フィルター内の磯砂をバケツに移し、手で平らにしてから、その表面まで水道水を加え、掻き混ぜて汚泥を剥離する。剥離汚泥を傾斜して、別の容器に移し、適宜、水道水で希釈・分別して、実験に使用する。
 硝化・脱窒菌が豊富で、また、硝酸イオンが蓄積しているので、汚泥の沈降速度などでは、測定中に汚泥が浮上する。循環ポンプを停止して、水道水をフィルターに注いで汚泥を洗浄するか、または、剥離汚泥に水道水を加えて、混合・沈降・傾斜を数回繰り返して、硝酸イオンを取り除くことにより、窒素発生による汚泥浮上を防止できる。

(3) Cultivation of experimental sludge

 A 60-liter aquarium set for ornamental fish (however, purification filter: 2 units, filter filler: Oiso sand 3-5 mm, bottom sand: Oiso sand 3-5 cm thick on the left) should be used. When the above skim milk powder is administered at 15 to 20 g/day, the purification filter is alternately regenerated at intervals of about 5 days, and about 1 L of good quality test sludge (MLSS 8,000 to 10,000 mg/L, SVI 50 mL/g ) is obtained.
 An appropriate dose of powder may be that the water quality is transparent at the time of powder administration on the next day. If in suspension, reduce the amount of powder dose. The water in the aquarium is replaced once a week with half of the aquarium water being replaced with tap water. For skim milk administration, no pH adjustment is necessary.
 To collect the sample sludge, when the purification filter overflows, move the sand in the filter to a bucket and flatten it by hand, then add tap water to the surface and stir to remove the sludge. The peeled sludge is tilted, transferred to another container, appropriately diluted with tap water, and used for the experiment.
 Since it is rich in nitrifying and denitrifying bacteria, and nitrate ions are accumulated, sludge floats up during measurement of settling velocity of sludge. Stop the circulation pump and pour tap water into the filter to wash the sludge, or add tap water to the exfoliated sludge and repeat mixing, settling and inclining several times to remove nitrate ions. Floating sludge due to nitrogen gas generation can be prevented.

7.供試汚泥の供給

 沈殿池・濃縮池・脱水機の設計・管理や凝集・脱水剤の種類選択・適正濃度・混合条件の決定など、実験・試験においては供試用の活性汚泥が必要となる。
 具体的には、稼働現場の活性汚泥を試験に用いることが最適であるが、試験する要員・場所・装置の関係で、遠隔な実験・研究室で実施することも少なくない。活性汚泥は、輸送・保存が困難であるので、実験室での培養を行うこととなる。室内実験での活性汚泥の育成・維持管理においては、様々なトラブルが生じる。トラブルの原因を解明し、適正な対応を実施できるようになるまでには、知識と経験が求められる。BOD負荷と酸素供給のバランスは、安定な維持管理にとって特別に重要である。
 回分・連続を問わず、初めて活性汚泥法を試みる際には、高価ではあるが、DO制御器の設置を勧める。図1に示す管理法は、簡単であるが、大学等の学生実験では勉強となるが、企業等では人件費等を考えるとDO制御システムを採用した方が結果的には低コストとなる。ただし、生物膜法においては、その必要はない。

7. Supply of sample sludge

 Activated sludge for testing is required in experiments and tests such as design and management of settling tank, concentration tank, dehydrator, coagulation, selection of dehydrating agent type, determination of proper concentration and mixing conditions.
 Specifically, it is best to use the activated sludge in the operation site for the test, but due to the personnel, place, and equipment of the test, it is often done in a remote experiment or laboratory. Since activated sludge is difficult to transport and store, it will be cultivated in the laboratory. Various problems occur in the growth and maintenance of activated sludge in laboratory experiments. Knowledge and experience are required before the cause of trouble can be clarified and appropriate measures can be taken. The balance between BOD load and oxygen supply is of special importance for stable maintenance.
 When trying the activated sludge method for the first time regardless of whether it is batch or continuous, it is recommended to install a DO controller although it is expensive. The equipment for management method shown in Fig.1 is simple, and it can be studied in student experiments at universities, etc. However, considering the personnel expenses etc. in companies, it is better to adopt the DO control system and will be lower cost. However, this control is not necessary in the biofilm method.

MLSS/MLVSS測定

 「JIS K 0102-14 懸濁物質及び蒸発残留物」に沿って測定する。
 ろ過器・乾燥機(MLSS:105~10℃)・電気炉(MLVSS:600℃)が必要である。

溶存酸素(DO)計

 電気化学式および光学式がある。価格も様々である。(a)出力端子を有し経時変化を記録計でモニターできるものか、または、(b)一定時間間隔で測定値を記憶し、CPUなどへデータ転送できるものがあれば、省力化が可能となる。
 (a)型はエアポンプに接続(リレー回路が必要)し、DOを所定値に維持できる。

pH計

 ガラス電極と参照電極の一体型を用いる。DO計と同様に、出力端子があれば、ペリスタポンプへ接続(リレー回路が必要)しアルカリ/酸溶液を注入して、pHを所定値に維持できる。

顕微鏡

 光学顕微鏡の中で生物顕微鏡(透過観察型顕微鏡)といわれるもの。安価な小学生科学実験用から、医学・生物の分野で用いられる高性能・高価なものまで、様々である。
 写真撮影やモニター付きがあれば、なおよい。写真撮影は、第三者の所見を求めることができる。モニターがあれば、グループで観察・ディスカッションができる。


Appendix – Major measuring equipments

MLSS/MLVSS measurement

 Measure according to “JIS K 0102-14 Suspended substances and evaporation residue” in Jpan.
 A filter, dryer (MLSS: 105-10°C) and electric furnace (MLVSS: 600°C) are required.

Dissolved oxygen (DO) meter

 There are electrochemical and optical types. Prices also vary. Labor saving is possible if there is (a) an output terminal that can monitor changes over time with a recorder, or (b) one that can store measured values ​​at a fixed time interval and transfer the data to a CPU, etc.
 Type (a) can be connected to an air pump (relay circuit is required) to maintain DO at a specified value.

pH meter

 An integrated type of glass electrode and reference electrode is used. As with the DO meter, if there is an output terminal, it can be connected to a peristaltic pump (requires a relay circuit) and injected with an alkaline/acid solution to maintain the pH at a specified value.

Microscope

 What is called a biological microscope (transmission observation type microscope) among optical microscopes. It ranges from inexpensive elementary school science experiments to high-performance and expensive ones used in the fields of medicine and biology.
 It would be even better if you had a photo shoot or a monitor. The photography may require the findings of a third party. If you have a monitor, you can observe and discuss in groups.
